Can the creation of an opportunity be restricted?

Meaning can I prevent standard users from creating an opportunity unless it's first a Lead?

Technically yes. It requires custom development though.

You have to override the Opportunity New Button so that a user cannot create the opportunity from the Opportunity Tab or Side Bar.

Then override the Convert button to allow the user to convert the lead into an Opportunity.
