Is there any way to append Two Fields (Source City, Destination City) values to one field, the reason for doing this is to enable Filter which allows City Selection to look for a City is either part of Source or Destination combination record
Since I have tens of Million records so I don't want to duplicate or Pivot the data

Did you mean create List parameter with manual entries?
if yes this will not solve the purpose, because source and destinations keeps adding or changing and mist of time new cities also added too
