Is it possible to add an action/trigger if a recipient replies to an Email?   

Engagement Studio - Reply to Email?

 ^ I currently have Email Open -> Assignment Status (but I'd rather this be if the recipient replies to an email).

    Nope, no trigger for replies. Pardot does not see the user's email inbox, and thus cannot take any actions.  There are various elaborate workarounds to track replies people have tried using  third party services like Zapier and salesforce linked inboxes, but nothing that can meaningfully impact your Engagement Studio Program.  Depending on the purpose of the program, one general strategy might be to use a dynamic suppression list for the program to stop the program if, for examples, someone was converted from a lead to a contact.
