That is the HUGE advantage of Permission Sets. You can associate 1 or more Users, from any Profile, to the Permission Set. Users can belong to multiple Permission Sets, so can have very basic permissions defined in Profiles (Users can only be in 1 Profile), and then layer on additional Permissions with Permission Sets -- adding Users as needed.
