Is there a simple way to import that list without producing duplicates? Or do I have to manually search all 100 of the leads to see if they're already in Salesforce?

No simple way to do this. There are tools out there like CRM Fusion's Demand tools that make this really simple, but it's costly. Other options are to do your own data scrubbing before loading by exporitng all of your leads to something like MS Access and running qieries to identify dupes.
