For Domain Registration Tool. Is there time reserved between once the "generate token" functionality becomes available and once this DNS record token is taken into consideration upon a send: after all, we need a bit of time to have it placed on the DNS server?

Yes there is a 90 day window before the token expires if not verified. The verification email token is only 48 hr window but the domain registration token provides 90 days
