For instance, is it possible for an Expired checkbox to appear as ticked (true) if the date is before today without having to modify a record? We would like the checkbox update to happen at 12:01am automatically.

If this could be a read-only field (ie, you can't change it separately from the Expiration date), then you can use a Formula Field.
In Summer '13, you'll be able to have a Checkbox Formula field, but until then, you have to use a Number or Text Formula, something like:
IF(TODAY() > Expiration_Date__c,"Expired","Active")
Then you could filter on that field.
