In a multi-user Salesforce instance, we'd like to be able to send out emails and have them come from (and reply to) the director, but have his assistant be able to send while logged into their account. This would require somehow changing the 'From' and 'Reply to' email address.
Is there a way to accomplish that in Salesforce?

@Claes Jonasson have you checked org wide email address? This will allow you to send email from a specific email address and it someone replies it goes to same email address.
Anyone in your org who has permission to use this email address can send from his behalf.
