If, as one of these Users, on the Opportunity, I click the "Add New" button on the related list for the custom object, I get a permissions error. Presumably, this is because I am doing it from the Opportunity.
But, if I go to the custom object's tab and create it from there, I can do so - including associating it to the Opportunity.
The custom object is always associated with an Opportunity, so creating it directly from the Opportunity makes 9000x more sense than doing it from the custom object's tab.
If you have set a User's profile to "Read Only" on an object (e.g. Opportunity), is there a way to enable them to add objects which they have "Read/Write" permissions on via a related list?

Thank you for your respone, Jeff. Org Wide Sharing is currently set to "Private". This seems correct, as the entire company shouldn't see this object, just the relevant profiles. The Profile in question here, under its settings, has Read/Write/Create/Delete rights to this object, with the exception of "View All" and "Modify All", which they shouldn't have. (They should only see the instance of the custom object that they are the owners of.)
