Hi, we are currently using LEX, but I am unable to use the Lightning Report builder or create new folders in Report or Dashboard tabs. Is there something in my org that needs to be enabled to use these new UI experiences?

Yup! These are all System Permissions that must be enabled on a Users Profile before a User can do them. "Report Builder (Lightning Experience)" is the System Permission required to use the Lightning Report Builder. And "Create Dashboard Folders" & "Create Report Folders" would be the others.
Just 1 word of caution. I believe the Lightning Report Builder is still in Beta... and I know that my experience so far as been a little rocky. Lots of crashing/resetting and having to start building my reports over again. You might want to stick with the Classic report builder a bit longer.
