Is it possible to see a preview of the email that will be sent to a donor / donors when the NPSP Email Acknowledgement button is pressed, either from within an Opportunity record, or from an Opportunity list view? And can it be editable (like a regular email message) so our ED can add some custom text if they wish?

Yeah, you're probably on the right track with the custom action. I haven't really had a chance to try from the standard one too much in LEX yet.
