When I instituted this rule it then required the field be entered no atter what stage was entered. I cannot see why that would be. Can someone put some fresh eyes to this and see if you can help?

Hi Shari...
You've got the ISBLANK( Win_Loss_Reason__c ) as part of your OR versus the AND... try this:
AND(OR(
ISPICKVAL( StageName , "80 - Verbally Awarded"),
ISPICKVAL ( StageName , "90 - PO in Process"),
ISPICKVAL ( StageName , "100 - PO Received"),
ISPICKVAL (StageName, "Lost Order")),
ISBLANK( Win_Loss_Reason__c )
)
