Upon an opportunity being closed/won, and any one of five fields has a value greater than 0, check to see if if any contacts on the related account contain a certain "Contact Role".
I suspect this is not possible, but wanted to check with the community.

Definitely not possible with the Validation rules.
You will need a custom code (apex) to validate the same.
