We cannot disable buttons on pagelayouts. We can hide them using record types and pagelyouts.
Other Option (Development):
Is it a custom button or standard button?
If it is a custom button we can show an alert, but we cannot disable the button.
If it's a standard button, we can override it and show a message that this record has already been compelted.
