We are transitioning to Lightning. All of our profiles are custom, except for Admin, which is standard. I would like to turn on Lightning just for one Admin in order for them to see and use the Switcher. I created a permission set and assigned this one Admin user to it. In the Lightning Transition Assistant, if I click on "Turn on Lightning Now" will that work? None of the custom profiles has the Lightning Enabled User checked, so they should not be affected, correct?

@Madeline Krazit, you are correct! You didn't need to assign the permission set to yourself (as the admin) as you have the permission through your Standard profile. As long as your users' profiles are custom and don't have the Lightning Experience User permission (and you haven't added them to the permission set), then they will be unaware of Lightning Experience when you turn it on in your org!
