How do I also make it so flow can capture this validation rule? I am getting errors that my flow has validation errors. In summary, I want it to be required that you fill out Expected Retailer Date if you select Yes in the picklist

Do you have the picklist field added to the screen as well? If so,try like this:
NOT(
AND
(ISBLANK ({!Expected_Retailer_Reflection_Date_On_sh__c}) ,
ISPICKVAL({!Know_Date_Retailer_Reflecting_Price__c} , 'Yes'))
)
Once you add the fields to the screen, click done and open the screen element again to add a validation rule in that screen.
