I'd like to set the creator user's manager on a custom sales supervisor field when registering an opportunity.
The custom sales supervisor field is in opportunity object.
I want to set it automatically. Is there any good way to set it?

Not sure why I didn't work for you. I created the same and it worked. You need to ensure that the manager field on user record is populated for the user who is going to create Opp record else it will be blank.
Step 2: On the action, please choose update a record from list and then in the value section, you need to follow Opportunity>>OwnerId>>Manager ID
