We want to make cases read only to one of our profiles after they have updated the status to closed while still leaving the case editable by the team leaders. is this possible?

One way to do this is to change the record type when the case status is closed to a different record type. That record type can be associated with a read only page layout for the one of four profiles you want to be read only. A second option is to use validation rules that speficially include that profile, and have criteria that look to see if the last modified date has changed
